What was tested

Rajasekar, Abdalla, Liang, Neelakantan and Yiu, the group behind this site’s 8 October analysis of their earlier peptide work, report in the International Endodontic Journal (published online 26 October 2026, ahead of print; DOI 10.1111/iej.70298; PMID 42929543) a step from dishes to animals for L803-mts, a myristoylated peptide that inhibits glycogen synthase kinase-3β and is used as a Wnt pathway agonist. In vitro, dental pulp stem cell monocultures and pulp stem cell-macrophage co-cultures were treated with L803-mts under an inflammatory challenge of 1 μg/mL lipopolysaccharide; odontogenic differentiation was read by alkaline phosphatase activity, gene expression, alizarin red staining and western blotting, and macrophage polarization by flow cytometry and enzyme-linked immunosorbent assay. In vivo, reparative dentinogenesis was measured in a rat pulp injury model by micro-computed tomography, histology and immunofluorescence, with mineral trioxide aggregate (MTA) as the reference capping material.

What they found

Under lipopolysaccharide exposure, L803-mts significantly increased mineralization and significantly upregulated the dentinogenic markers dentine matrix acidic phosphoprotein 1 (DMP1) and dentine sialophosphoprotein (DSPP). In co-culture, it significantly reduced pro-inflammatory cytokines and shifted macrophages toward the anti-inflammatory phenotype (p < 0.05). In the rat model, the peptide achieved mineral density comparable to MTA (1.17 versus 1.32 gHA/cm³, p > 0.05) but significantly lower mineral volume (0.03 versus 0.06 mm³, p < 0.05). The authors conclude that L803-mts promoted reparative dentinogenesis through Wnt signaling activation and immunomodulation.

What it does not show

This record sits at the level of the author-supplied abstract; the paywalled full text was not accessible, so no figure-level numbers, group sizes or dose ranges are quoted here. Three boundaries hold even at that level. First, the parity claim concerns mineral density only: total mineral volume was half that of MTA and significantly lower, so “comparable to MTA” is not the whole in vivo result. Second, the model is a rat pulp injury, not carious pulpitis in a patient, and the abstract reports no functional, vascular or long-term outcomes beyond the imaging and histology endpoints listed. Third, the Wnt attribution is the authors’ interpretation; the abstract-level record contains no rescue or loss-of-function experiment, so the signaling mechanism remains asserted rather than demonstrated, the same gap flagged in this group’s in vitro paper.

Where it sits in the field

For the pulp and dentine repair program, this is the first in vivo entry in the peptide GSK-3β lane this site tracks. The 8 October piece recorded exactly the gaps this study begins to close: no inflammatory environment, no immune compartment, no animal. All three now exist, in the same journal that recently carried the group’s calcium strontium silicate pulpotomy study in rat molars, which suggests the in vivo side of the program is consolidating. It does not move any program tier: one rat model, abstract-level reporting and a volume deficit against MTA keep this a bounded proof of progress rather than a parity claim with the clinical standard. The current field assessment stands at /field/.

Where we differ from the coverage

We found no press or popular coverage of this paper to differ from. Against the authors’ own conclusion, the framing “through Wnt signaling activation and immunomodulation” is a hypothesis the abstract-level data are consistent with but do not isolate, and “comparable to MTA” describes mineral density only, not the amount of dentine formed. We state both boundaries rather than the headline version.
